In this chapter we review the three-dimensional structural analysis of axonemal dynein and other proteins of eukaryotic flagella and cilia in situ as revealed by electron tomography [5, 6, 20, 24, 31, 39, 41, 42, 47] and electron microscopy [7, 44, 59]. We review historical landmark contributions to the field and technical details of electron tomography and present an outlook for future developments.
